TP-FLIM双光子荧光寿命成像光学造影剂

双光子荧光寿命成像(Two-photon fluorescence lifetime imaging, TP-FLIM)技术集合了双光子荧光成像和荧光寿命成像的优势,能够对生物组织进行高分辨三维成像的同时提供生物组织的生化特性信息,在**诊疗方面具有巨大的临床应用前景。为了获得佳的双光子荧光寿命成像效果,光学造影剂除了需要具有较强的双光子吸收,用于降低激光的能量,避免高能量激光对组织的损伤;还需要具有稳定且较长的荧光寿命,用于去除生物自荧光对双光子荧光寿命成像的干扰。然而,目前还鲜有材料能够同时满足上述要求。因此,迫切需要设计合成一种合适光学造影剂,用于推动双光子荧光寿命成像技术在生物诊疗领域的发展。

一种基于热激活延迟荧光(Thermally activated delayed uorescence, TADF)的有机光学造影剂,可用于双光子活体荧光寿命成像。在该工作中,研究者充分利用热激活延迟荧光材料通常具有较长延迟荧光(Delayed fluorescence)寿命这一优点,将具有聚集诱导荧光增强的有机热激活延迟荧光材料包覆于两亲性共聚物内,成功制备了水溶性的有机纳米颗粒。光学研究表明两亲性共聚物的包覆大化了热激活延迟荧光材料的聚集态,大大增加了材料的荧光寿命,获得了可媲美磷光材料的超长发光寿命(4.2 μs)。更为重要的是,不像传统磷光发光强度和寿命容易被氧气猝灭,该材料对氧气表现出明显的惰性,即便在富氧区域仍然表现出稳定的荧光寿命,非常有利于获得高信噪比的荧光寿命成像。同时,该热激活延迟荧光材料由于具有Donor-AcceptorD-A)结构,表现出良好的双光子吸收截面,轻易实现了活体双光子荧光寿命成像。

该研究为双光子荧光寿命成像技术提供了一种良好的光学造影剂,对推动双光子荧光寿命成像技术在临床**诊疗中的发展具有重要意义。
