DBCO (dibenzocyclic octyne) polyethylene glycol mercaptan (DBCO-PEG-SH) derivatives can conduct chemical reactions without any metal catalyst. The 1,3-dipolar cycloaddition reaction of cyclooctyne and azide compounds promoted by the strain, also known as copper free click reaction, is a bioorthogonal reaction, which can make two molecules conjugate in aqueous solution. DBCO-PEG derivatives have fast kinetics and stability in aqueous buffer solution. DBCO reagent can be used to label azide modified biomolecules without toxic copper catalyst. Polyethylene glycol can increase the solubility and stability of peptides and proteins, and reduce their immunogenicity. It can also inhibit the nonspecific binding of charged molecules to the modified surface.
