外泌体是细胞分泌的细胞外囊泡,其通常在40-150 nm范围内。外泌体具有调节多种生理过程的功能,并且可能和一些疾病相关,但是目前对外泌体的研究仅停留在其糖基化的整体状态上,而对于单独蛋白上外泌体的糖特征的研究还没有合适的工具。
开发了一种叫做定量局部分析(QLA)的方法来对外泌体上的蛋白质特意糖信号进行研究,这个方法的构建**是分离了MCF-7细胞的外泌体,并对其进行表面构架:向其中加入靶向上皮细胞表达的扩膜蛋白EpCAM的适配体A,并且将适配体用6-巯基-1-己醇进行修饰,从而通过6-巯基-1-己醇将外泌体固定在金电极上,之后向其中加入靶向MUC1的适配体B,并且适配体B同时与失活的半乳糖氧化酶连接。这里MUC1是外泌体黏蛋白1,在外泌体中大量表达,而失活的半乳糖氧化酶是加入了[Fe(CN)6]4-的,而这个过程中加入Cu2+可以通过与[Fe(CN)6]4-形成六氰合铁酸铜(CuHCF),从而使半乳糖氧化酶活性恢复。之后在半乳糖氧化酶活性恢复后,可以使其附近的外泌体上糖修饰的末端半乳糖氧化,从而产生醛基,与生物素酰肼进行生物正交反应,并于链霉素-辣根过氧化物酶复合物结合,而辣根过氧化物酶在六氰合铁酸铜与对苯二酚存在下,可以产生电子流,这个可以被金电极通过循环伏安法以及差动脉冲伏安法检测到,而信号的强度取决于外泌体上连接的辣根过氧化酶数目,也就是说与外泌体上的糖修饰相关,因此可以进行后续的对外泌体的检测。
